Isozymes from avocado cotyledons

Abstract

Liquid nitrogen and a protectant buffer were used to prepare extracts from the cotyledons of avocado (Persea americana Mill.) for isozyme analysis with electrophoresis. Zymograms of several cotyledon enzyme systems are the same as those of mesocarp, and leaves and several systems can be clearly resolved for at least 180 days following the onset of germination. Screening for pollen parentage in breeding programs can now be done before planting out since the assays are nondestructive and a given cotyledon can be assayed repeatedly.
