Hypotonic bursting method for spreading synaptonemal complexes of Zea mays

Abstract
An adaptation of the hypotonic bursting technique for spreading synaptonemal complexes (SCs) is described for primary microsporocytes of Zea mays. SCs are spread in two dimensions and stained with silver for observations in the light or electron microscope. Substages of pachytene can be determined accurately using changes in the structure of the nucleolus. This technique allows detailed analyses of the synaptic pattern in complete sets of SCs, including synaptic irregularities such as inversion loops.
