The nucleolus organizer chromosomes of Nicotiana tabacum

Abstract
Nucleolar organizer regions (NORs) could potentially serve as good chromosome markers for Nicotiana tabacum. However, the number of NOR chromosomes in N. tabacum has been disputed, with reports ranging from one to three pairs. The objective of this study was to accurately determine the number of NORs for this species. Data were collected both on number of satellited chromosomes observed in mitotic metaphases and on association of NORs with nucleoll in meiotic diplotene preparations. Several cultivars were included to determine if variability for number of NORs exists within the species. All cultivars studied had a maximum of three bivalents attached to nucleoli during diplotene. Six satellited chromosomes were observed in mitotic metaphases of cvs. Burley 21 and NC95. Only four satellited chromosomes were regularly observed in cvs. SC58 and Coker 139; however, three types of satellited chromosomes appeared to be present in the former and a third pair of satellited chromosomes was observed in mitotic prophase of the latter cultivar. Therefore, although the mitotic metaphase data were not conclusive for two of the cultivars studied, it appears that N. tabacum has three pairs of satellited chromosomes and that all of these organize nucleoll.
