Identification of D-Genome chromosomes of chinese spring of sourthern Blots using Gnome-specific repetative DNA

Abstract

Two D-genome-specific repetitive DNA probes were hybridized to restricted DNA from D-genome disomic substitution lines. These lines contained a single D-genome chromosome substituted for its A or B genome homoeologue in Langdon durum wheat. Both probes hybridized to DNA on every D-genome chromosome. One of the probes (pAS1) revealed a unique hybridization pattern for each chromosome using a single restriction enzyme, and thus it may be useful for the construction and monitoring of D-genome aneuploid wheat stocks.
