Isolation and Genetic Analysis of a Triazolopyrimidine-Resistant Mutant of Arabidopsis

Abstract
We have isolated a triazolopyrimidine (herbicide)-resistant mutant by screening seedlings of M2 populations from EMS-treated seed of Arabidopsis thaliana Columbia wild type. The resistance was found to be due to a single, dominant, nuclear gene mutation. The mutant gene maps to linkage group 3 and is allelic to the CSR1 locus that encodes the acetolactate synthase (ALS) enzyme. At the plant growth level the mutant is about 500-fold more resistant to triazolopyrimidine than is the wild type. ALS activity in enzyme extracts from the mutant was about 1,000-fold less sensitive to inhibition by triazolopyrimidine than in extracts from wild-type plants. Resistance of whole plants to triazolopyrimidine and differences in inhibition of ALS activity cosegregated, indicating that plant and enzymatic resistances were caused by the same mutant allele at the CSR1 locus. The new mutant allele was denoted csr1-3.
