Abstract
Spontaneous male-sterile, female-fertile and male-sterile, female-sterile mutations in soybean, Glycine max (L.) Merr. were analyzed to determine whether they represent new or already described loci responsible for sterility. Cytological abnormalities of microspores revealed a variety of conditions existing in sterile anthers from male-sterile, female-fertile mutations: coenocytic microspores in T287 and T290 mutants and degenerated debris in sterile anthers in the T291 mutant. Diversity in microspore appearance was observed in sterile T292 plants. On the basis of allelism tests, the T287 and T290 mutants were associated with the ms1 locus and were designed ms1 (Ames 2) and ms 1 (Danbury), respectively. The T291 mutant was located at the ms3 locus and designated ms3 (Plainview), whereas the T292 mutant was located at the ms4 locus and designated ms4 (Fischer). Two independent mutations, ISU-sterile and Beeson-sterile, represented new sources of alleles at st2 and st5 loci, respectively. Linkage tests involving the Beeson-sterile mutation showed 24.7 ± 4.3% recombination with the w1 locus and provided additional evidence that this mutation and the st5 mutation are controlled by alleles at the same locus.
